I've been told that if a FCU has a plenum with four spigots on (two on the front and one each side) and I only require to use two of them, then I should use the front two even if the grilles I am serving are at the side (or even behind) the unit. Something to do with resistance...
Is this correct?

Right. This has been a difficult one and we've contacted several FCU manufacturers.
It would appear that there are several issues involved including the number and location of fans within the unit, size and shape of plenum box, etc. and (judging by the response from manufacturers) it would be wise to contact them and seek their recommendations as these do differ.
Perhaps it's important to make a selection of preferred supplier/manufacturer early in the contract to avoid abortive draughting and co-ordination time (if this issue is taken on board and then found not to be required, or vice- versa) or possible additional material costs if the ductwork is routed to accomodate this issue and then found not to be required.

Anyone else have a view on this?

I would check what the static pressure is of the fan & if the sytem resistance is well under what the fan can do then go the cheaper option.
